Characterization and phylogenetic analysis of chloroplast genome of Berberis dasystachya Maxim
[Objective]The aim of this study is to enrich the data of chloroplast genome in Berberisand provide a reference for further research on genetic resources,species identification and phylogenetic analysis of Berberisand dasystachya.[Method]Based on high-throughput sequencing technology,the complete chloroplast genome sequence of B.dasystachya was obtained by bioinformatics ap-proaches.Next,its simple sequence repeat,codon preference and IR region expansion and contraction were analyzed.The phyloge-netic analysis of B.dasystachya and its related groups was carried out.[Result]The chloroplast genome of B.dasystachya was 167 036 bp in length with GC content of 38.16%,and encoded 144 functional proteins.Up to 109 simple sequence repeat(SSR)loci were detected in the chloroplast genome,and most of them(73.4%)were single-nucleotide repeats composed of A or T.Phylo-genetic analysis showed that B.dasystachya had the closest genetic relationship with B.koreana,B.amurensis and B.weiningensis,and was located in the same clade with Mahonia oiwakensis and Mahonia bealei.[Conclusion]Analyze sequence characteristics of the chloroplast genome can support the phylogenetic relationship of Berberidaceae to a certain extent.
