短短芽胞杆菌分支酸合成酶基因 BbaroC的鉴定及转录因子调控分析

分支酸合成酶AroC是莽草酸途径中芳香族化合物合成的关键酶,探明短短芽胞杆菌aroC基因的特征及转录调控因子可为短短芽胞杆菌作用机理研究奠定基础。从短短芽胞杆菌FJAT-0809-GLX中克隆了 aroC基因,进行生物信息学分析和转录因子预测。短短芽胞杆菌FJAT-0809-GLX中aroC基因序列长度为1164 bp,GenBank登录号为OR475562。短短芽胞杆菌FJAT-0809-GLX 与 Brevibacillus brevis NBRC 100599 的 aroC 基因序列同源性最高,为 98。88%,与Brevibacillus brevis HNCS-1的AroC氨基酸序列同源性最高,为100%;其编码的蛋白为亲水性蛋白,无信号肽,定位于细胞质。靶向BbaroC的转录因子共有23个,分别来自10个物种,Escherichia coli(Strain K12 MG1655)和 Bacillus subtilis(Strain 168)分别预测到 12 个和 3 个转录因子。BbaroC在短短芽胞杆菌抑菌活性物质产生中可能发挥着重要作用,这为其在农业上的广泛应用提供依据。
Identification of the Chorismic Acid Synthetase Gene BbaroC in Brevibacillus brevis and Analysis of the Transcription Factor Regulation
The chorismic acid synthetase AroC is the key enzyme for the synthesis of aromatic compounds in the shikimic acid pathway.Identifying the characteristics of aroC gene in Brevibacillus brevis and its transcriptional regu-lators will lay a foundation for the study of the mechanism of action of Brevibacillus brevis.The aroC gene was cloned from Brevibacillus brevis FJAT-0809-GLX,and then the bioinformatics analysis and the prediction of transcription factor were carried out.The sequence length of aroC gene in Brevibacillus brevis FJ AT-0809-GLX was 1 164 bp,and the GenBank accession number was OR475562.The aroC gene sequence of Brevibacillus brevis FJAT-0809-GLX had the highest homology with that of Brevibacillus brevis NBRC 100599,which was 98.88%.The amino acid sequence of AroC had the highest homology with Brevibacillus brevis HNCS-1,which was 100%.Its encoded protein was a hydrophilic protein without signal peptide,which was located in the cytoplasm.A total of 23 transcription factors tar-geting BbaroC were identified from 10 species,with 12 and 3 transcription factors predicted from Escherichia coli(Strain K12 MG1655)and Bacillus subtilis(Strain 168),respectively.BbaroC may play an important role in the pro-duction of antibacterial active substances of Brevibacillus brevis,which would provide basis for its wide application in agriculture.
