胧月总黄酮提取工艺优化

Optimization of Extraction Technology of Total Flavonoids in Graptopetalum paraguayense

以胧月叶片为原料提取总黄酮,比较甲醇提取法、乙醇提取法、超声辅助-乙醇提取法的提取效果,最终选择超声辅助-乙醇提取法为最佳提取方法.利用单因素试验和响应面法对提取工艺参数进行优化,以黄酮提取率为考察指标,确定最佳提取条件为超声时间41 min、超声温度54 ℃、液料比16∶1(mL∶g)、乙醇体积分数85%、超声功率250 W.在此工艺条件下,测得胧月总黄酮的最大提取率为 2.4%.
Flavonoids were extracted from Graptopetalum paraguayense leaves via ultrasonic-assisted etha-nol extraction,which exhibited significantly higher yields in comparison with traditional methods involving methanol and ethanol extraction.The main factors were investigated by one factor test and response sur-face methodology.The results showed that the optimal conditions were as follows:ultrasonic time 41 min,temperature 54 ℃,liquid-solid ratio 16∶1(mL∶g),ethanol volume fraction 85%and ultrasonic power 250 W.Under these settings,the yield reached 2.4%.
