Targeted Protein Degradation via Genetically Encoded Degraders
Genetically encoded targeted protein degradation(GE-TPD)is a therapeutic ap-proach that leverages genetic information to express degrading molecules within cells.It currently plays a crucial role in the degradation of previously"undruggable"proteins associated with various cancers and other diseases.This review summarizes recent advancements in GE-TPD,including the design and selection of E3 ligases and target protein binding domains,as well as module selection,and explores the prospects and challenges of its personalized applications.
