摘要
基因编码的靶向蛋白降解是一种以遗传信息为基础的降解分子在细胞内表达编码的治疗方法,已经在许多癌症疾病的"不可药用"蛋白的降解中发挥着极为重要的作用.综述了GE-TPD近年来的一些进展及其E3 连接酶和目标蛋白结合域的设计和模块选择;并展望了GE-TPD个性化定制的前景和挑战.
Abstract
Genetically encoded targeted protein degradation(GE-TPD)is a therapeutic ap-proach that leverages genetic information to express degrading molecules within cells.It currently plays a crucial role in the degradation of previously"undruggable"proteins associated with various cancers and other diseases.This review summarizes recent advancements in GE-TPD,including the design and selection of E3 ligases and target protein binding domains,as well as module selection,and explores the prospects and challenges of its personalized applications.
