基于CTAB改性凹土的细菌基因组DNA提取方法研究

摘要
利用HCl和十六烷基三甲基溴化铵(CTAB)对凹凸棒土进行改性,增加其表面功能基团,研究改性材料对脱氧核糖核酸(DNA)的吸附与解吸效果.通过X射线衍射(XRD)仪、傅里叶变换红外光谱(FTIR)仪、扫描电子显微镜(SEM)和比表面积测试仪(BET)等对改性前后的凹凸棒土进行表征.以金黄色葡萄球菌为例,考察改性凹凸棒土用于核酸提取的得量、纯度及完整性.结果表明,改性材料对DNA的饱和吸附量可达18.657 μg/mg,对金黄色葡萄球菌核酸提取得量为(5.45±0.46)ng/105CFU,且结构完整、纯度高.
Abstract
Attapulgite was modified with HCl and CTAB to increase functional groups on the surface of attapulgite,and the adsorption and de-sorption of modified materials on DNA were studied.X-ray diffractometer(XRD),Fourier transform infrared spectroscopy(FTIR)and scanning elec-tron microscope(SEM)were used to characterize attapulgite before and after modification.The amount,purity and integrity of nucleic acid extraction were investigated with Staphylococcus aureus as the object of nucleic acid extraction.The results showed that the saturated adsorption capacity of the modified material for DNA could reach 18.657 μg/mg,the nucleic acid extraction capacity of Staphylococcus aureus was(5.45±0.46)ng/105 CFU,the structure was intact and the purity was high.
