The Expression and Correlation of Serum MMP-7 and ADAM17 in Connective Tissue Disease-related Interstitial Lung Disease
Objective:To investigate the expression level and correlation of matrix metalloproteinase-7(MMP-7)and a disintegrin and metalloproteinase 17(ADAM17)in connective tissue disease(CTD)-related interstitial lung disease(ILD).Methods:A total of 40 CTD-ILD patients treated in the Rheumatology and Immunology Department of the First and Second Affiliated Hospital of Baotou Medical College from December 2021 to July 2023 were collected as the CTD-ILD group,30 CTD patients without ILD as the CTD group,and 26 healthy individuals who underwent physical examinations during the same period as the control group.Basic information of patients was collected,and the expression levels of MMP-7 and ADAM17 in serum were detected by enzyme-linked immunosorbent assay(ELISA)in the three groups;CTD-ILD was classified into the mild,the moderate,and the severe according to the percentage of carbon monoxide diffusion to the expected value[DLCO(%pred)].The expression levels of serum MMP-7 and ADAM17 in different subgroups of CTD-ILD and lung function groups were analyzed,and the correlation between MMP-7 and ADAM17 with various indicators was elucidated,as well as their value in diagnosing CTD-ILD.Results:① Compared with the CTD group and the control group,the serum MMP-7 and ADAM17 levels in the CTD-ILD group were significantly increased(P<0.01);② In different subgroups of CTD-ILD,compared with the SS-ILD group,serum MMP-7 and ADAM17 levels were higher in the SSc-ILD group(P<0.05);③ In different lung function groups of CTD-ILD,serum MMP-7 levels were as follows:the severe was greater than the moderate,and the moderate was greater than the mild(P<0.01),and serum ADAM17 levels were higher in the moderate and severe groups than in the mild group(P<0.01);④ Correlation analysis showed that MMP-7 was positively correlated with CT score,CPI,and age in CTD-ILD patients(P<0.05),ADAM17 was positively correlated with CT score and CPI(P<0.01),and MMP-7 and ADAM17 were negatively correlated with FEV1,FVC,DLCO(%pred),and P(O2)(P<0.05).Conclusion:Serum MMP-7 and ADAM17 may be involved in the occurrence and development of CTD-ILD,and have certain predictive value for evaluating the severity of CTD-ILD.
