摘要
目的:揭示慈溪市汉族脑外伤患者内脂素基因-1535 C>T位点单核苷酸多态性与血浆内脂素及C-反应蛋白(CRP)水平的相关性.方法:选取98例重型颅脑损伤患者和98例年龄及性别均配对的体检健康者,采用聚合酶反应-限制性片断多态性分析基因型,同时采用ELISA检测血浆内脂素及CRP浓度,统计分析其相关性.结果:酶切后可见内脂素基因-1535 C>T位点有TT、TC和CC三种基因型.经卡方检验,脑外伤患者及正常对照组各基因型分布及等位基因频率比较差异均无统计学意义(均P>0.05).经方差分析,CC基因型患者血浆内脂素或CRP浓度均显著高于CT和TT基因型患者(均P<0.01),而CT和TT基因型患者血浆内脂素或CRP浓度差异均无统计学意义(均P>0.05).Pearson相关分析显示,血浆CRP浓度与血浆内脂素浓度呈显著正相关(P<0.01).结论:慈溪市汉族脑外伤患者内脂素基因-1535 C>T位点单核苷酸多态性与血浆内脂素和CRP浓度显著相关,可能参与外伤性脑损伤的炎症反应.
Abstract
Objective This study aimed to examine the correlativity of the Visfatin gene-1535 C > T single nucleotide polymorphism of visfatin gene to the plasma visfatin and C-reactive protein( CRP) levels in Han patients with traumatic brain injury in Cixi cit-y. Methods 98 patients with sevens traumatic brain injury and 98 age- and sex-matched healthy subjects were recruited. Genotypes were analyzed using FCR-restriction fragment length polymorphism. Plasma visfatin and CRP concentrations were measured with ELISA. And, furthermore, their relationships were analyzed statistically. Results The genotypes of the -1535 C>T polymorphism of visfatin gene included TT, CT and CC. The genotype and allele frequencies between healthy subjects and patients showed no significant differences (all P>0.05) using chi-square test. Using analysis of variance, genotype CC presented distinctly plasma visfatin and CRP concentrations (all P<0.01) ; the plasma visfatin and CRP concentrations between genotype CT and TT manifested no significant difference (all P>0.05). Plasma visfatin level expressed significantly associated with plasma CRP level (P <0.01) using pearson correlation test. Conclusion The -1535 C >T single nucleotide polymorphism of visfatin gene was associated with plasma visfatin and CRP concentrations and seemed to be potentially involved in the inflammatory component of traumatic brain injury in Han patients with head trauma in Cixi city.
