摘要
目的:探讨缺氧诱导因子-1a(HIF-1a)和血管内皮生长因子(VEGF)蛋白在结直肠癌组织中的表达及与结直肠癌临床病理特征的相关性.方法:采用免疫组化SP法检测39例结直肠癌组织、39例相应癌旁胃组织(距癌组织边缘>5cm)中HIF-1a和VEGF蛋白的表达,分析结直肠癌组织中HIF-1a和VEGF蛋白表达与结直肠癌临床病理特征的相关性,并分析HIF-1a和VEGF蛋白表达之间是否相关.结果:癌旁胃组织中HIF-1a无表达,VEGF蛋白低表达12.8%(5/39),癌组织中HIF-1a和VEGF蛋白阳性表达率分别为61.5%(24/39)和66.7%(26/39),与癌旁组织相比均差异显著(P<0.05);HIF-1a和VEGF蛋白表达与结直肠癌TNM分期、浸润深度及是否淋巴结转移密切相关(P<0.05),而与性别、年龄、肿瘤直径和分化程度无关;癌组织中HIF-1a与VEGF蛋白表达呈正相关(r=0.374,P<0.05).结论:HIF-1a和VEGF蛋白在癌组织中表达较癌旁组织显著增高表明其可能与结直肠癌的发生、发展及淋巴结转移有关.联检HIF-1a和VEGF蛋白的表达,有利于评估结直肠癌的生物学行为,提高预后判断的准确性,并对临床治疗有重要指导意义.
Abstract
Objective To investigate the expression of HIF-1a and VEGF protein in tissues of colorectal carcinoma and its correlation with clinicopathological features. Methods The expression of HIF-1a and VEGF protein in 39 tissues of colorectal carcinoma and 39 adjacent gastric tissues (the distance from the edge of the cancerous tissue is >5cm) were detected by immunohistochemistry, and the correlation between the expression of HIF-1a and VEGF protein in the tissues of colorectal malignancies and its association with clinicopathological factors were analyzed. Results There was no positive expression of HIF-1a in adjacent gastric tissues. The positive expression rate of HIF-1a protein in tissues of colorectal carcinoma was 61.5% (24/39) ,and that of VEGF protein was 66.7% (26/ 39). The expression of HIF-1a and VEGF protein showed closely related to lymph node metastasis,tumor stage and invasive depth (P < 0. 05) ,but no relativity to gender,age,tumor diameter and differentiation. The expression of HIF-1a protein in tissues was positive correlative with VEGF protein( r = 0. 374, P < 0. 05 ). Conclusion The expression of HIF-1a and VEGF protein in cancerous tissues manifest higher significantly than adjacent tissues. It is probably related to the carcinogenesis,development and lymph node metastasis. Combined analysis of the expression of HIF-1a and VEGF protein has benefit in evaluation of the biological behaviour of colorectal carcinoma .increases the accuracy of prognostication and plays an important guiding role in the clinical treatment.
