Objective To explore the method radiolabeled monoclonal antibody 4E5 with radionuclide 125I, and to observe the biological distribution of 125I-4E5 in normal mice. Methods 4ES was radiolabeled with 125I by the Iodogen method at room temperature . The labeling rate and radiochemical purity was tested with TCA, and the stability of 125I-4E5 was analyzed. 43 mice were divided into nine groups randomly, and 125I-4E5 was injected in the tail vein with 148KBq/0.2 ml separately. The biodistribution of 125I-4E5 was measured at 5min, 15min, 30 min, 1 h, 2 h, 6 h, 24 h, 48 h and 72 h after injection. Results The labeling rate of 125I-4E5 proved(79.2 ±2.6)% , and the radiochemical purity showed(97.1 ±1.1)% . Its specific activity expressed 294.5MBq/mg. The radiochemical purity of 125I-4E5 mixing with serm and PBS indicated over 90% after one week. The body distribution shows 125I-4E5 was deposited mainly in liver, spleen and kidneys, and the blood pool presented 125I-4E5 was eliminated quickly. Conclusion The labeling rate and radiochemical purity of 125I-4E5 using the Iodogen method is high, simple and the stability of 125I-4E5 is fine. The metabolism of 125I-4E5 in normal mice mainly depend on liver and kidneys and with fast clearance in blood.
