目的:建立三碘甲腺原氨酸(T3)第三次定量免疫测定用国家标准品.方法和结果:精确称取高纯T3原料,用含0.5%BSA的0.02M PBS缓冲液配制成100ng/ml溶液,经无菌过滤处理后,分装、冻干制成候选品.以T3第二次国家标准品(150550-0004)为对照品,经协作标定,候选品中T3免疫活性为50ng/支.结论:候选品各项指标均符合要求,可作为T3第三次免疫测定用国家标准品使用.本标准品的建立,为T3免疫测定用试剂盒的标准化提供定量基础.
Abstract
Objective To establish the 3rd of national standardized products of quantitative immunoassay of 3, 5, 3'-Triiodothy ronine(T3 )in China. Method and Result High purity of T3 raw material were precisely weighted to prepare 100ng/ml solution with 0.02M PBS buffer containing 0.5% BSA, then sterile filtered, filled and lyophilized to prepare the national standard candidate. The candidate was calibrated collaboratively using 2nd national standard as standards(150550-0004). The immune activity of candidate was 50ng/ampoule. Conclusion The candidate met the relevant requirements and can be served as the 3rd of national standard for quantitative immunoassay of T3. The establishment of the 3rd of national standard for quantitative immunoassay of T3 will provide a quantitative basis for quantitative immunoassay kits of T3.
