Objective:To investigate iron accumulation and its relationship with depressive symptoms in the limbic system and basal ganglia of patients with major depressive disorder(MDD)and bipolar disorder(BD)using quantitative susceptibility mapping(QSM).Methods:Whole brain QSM images and Hamilton Depression Scale(HAMD)scores of the subjects were collected.Regions of interest(ROI)including the caudate nucleus,lateral globus pallidus,medial globus pallidus,puta-men,substantia nigra,subthalamic nucleus,gyrus rectus,medial orbital gyrus,posterior orbital gyrus,fusiform gyrus,parahippocampal gyrus,superior temporal gyrus,middle temporal gyrus,and inferior temporal gyrus were selected.Mean susceptibility values and laterality index(LI)were calculated for each ROI.The Kruskal-Wallis test for multiple independent samples was used to compare between groups.Spearman correlation analysis was used to examine the correlation between susceptibility val-ues and HAMD scores.Results:Compared with the BD group,the MDD group had a significantly lower susceptibility value in the left parahippocampal gyrus(P=0.047).The LI of the medial orbital gyrus in the MDD group was higher than that in BD group(P=0.005).Correlation analysis revealed a posi-tive correlation between HAMD scores and susceptibility values in the right caudate nucleus in the MDD group(r=0.569,P=0.009),as well as between susceptibility values in the right middle tempo-ral gyrus in the BD group(r=0.545,P=0.013).Conclusions:There are regional and laterality differ-ences in iron deposition in patients with MDD and BD.The level of iron deposition in specific brain re-gions is associated with the severity of depressive symptoms.
