摘要

从采集的滨海盐土样品中分离出多株放线菌,通过滤纸片法从中筛选具有抗菌活性的菌株,对活性菌株进行鉴定和发酵条件优化.实验筛选得到可产生不同抗菌活性物质的3株链霉菌,其中菌株ZJPH2021031的发酵液对金黄色葡萄球菌呈现出较质量浓度为50 mmol/L的硫酸链霉素更高的抑制活性;菌株ZJPH2021032的发酵液对革兰氏阳性菌和真菌的生长具有抑制作用;菌株ZJPH2021033的发酵液则仅对真菌表现出抑制活性.采用含多种金属离子和以氨基酸为氮源的培养基发酵时,菌株ZJPH2021031的发酵液对金黄色葡萄球菌的抑制活性得到显著提升,抑菌圈直径增大了2.16倍,且培养基中的甘油质量浓度对抗菌活性物质产生显著的影响.

Abstract

The filter paper method was used to screen strains with antimicrobial activity,and the active strains were identified,the optimization of fermentation conditions was also conducted . The results showed that three strains of Streptomyces with different antibacterial activities were obtained,among which the fermentation broth of strain ZJPH2021031 showed higher inhibitory activity against Staphylococcus aureus than that of streptomycin sulfate at a concentration of 50 mmol/L .The fermentation broth of strain ZJPH2021032 inhibits the growth of Gram-positive bacteria and fungi,while the fermentation broth of ZJPH2021033 only inhibited the growth of fungi .Meanwhile,the inhibitory activity of the fermentation broth of strain ZJPH2021031 against Staphylococcus aureus was significantly enhanced,and the bacteriostatic diameter increased by 2 .16-fold w hen fermented in a medium containing various metal ions and amino acids only as a nitrogen source . Additionally,the glycerol content of the medium was the most important for the production of antimicrobial active substances .
