固定化乳酸菌生产乳酸工艺条件的优化

Optimization of the process conditions for lactic acid production by immobilized lactic acid bacterium

作为一种天然有机酸,乳酸广泛应用于医药、食品、农业、工业及生物材料等领域.为满足乳酸市场需求、降低生产成本,优化乳酸生产条件、筛选高产乳酸菌株并应用于工业生产已成为当前研究的重点.利用实验室诱变育种得到高产乳酸菌株LBZ2,利用该菌株进行固定化乳酸发酵,开展单因素实验,结合响应面法优化固定化生产乳酸的条件,比较LBZ2液体发酵与固定化包埋的发酵产酸能力.研究结果表明:乳酸菌株LBZ2固定化发酵产酸量较高,其最佳产酸条件为K型卡拉胶质量分数2.3%,LBZ2接种量15.3%,固化液KC1质量分数3.0%,发酵时间48 h,在该条件下乳酸产量达84.66 g/L,体积产率为1.763 g/(L·h).
As a natural organic acid,lactic acid is widely used in medicine,food,agriculture,industry and,biomaterials.To meet the market demand for lactic acid,research on production cost reduction,production condition optimization,high-yield lactic acid strain screening and their application in industrial production of lactic acid has become a hotpoint.The high-yielding lactic acid strain LBZ2 was obtained through laboratory mutagenesis breeding,and this strain was used to perform immobilized lactic acid fermentation.A single factor experiment was conducted to optimize the conditions for immobilized lactic acid production using the response surface method,and the acid production capacity of LBZ2 liquid fermentation and immobilized embedding was compared.The results showed that the acid yield of lactic acid strain LBZ2 was high after immobilized fermentation,and the optimal acid production conditions were 2.3%for K-type carrageenan,15.3%for LBZ2 inoculation,3.0%for KCl in solidifying solution,and 48 h for fermentation time,under which the lactic acid yield was 84.66 g/L and the volume yield was 1.763 g/(L·h).
