Identification and Biological Characteristics of the Pathogens Causing Root Rot Disease of Artocarpus heteophyllus Lam in Hainan Province
The disease roots of A rtocarpus heteophyllus Lam with root rot symptom were sampled in Hainan,China.Sixty-two fungal isolates were finally from 15 samples ofjackfruit root rot collected from 11 cities. The pathogen of the predominant fungus from the samples of diseased tissues was tested according to Koch's law,the Pathogen was identified according to morphological Characteristics and ribosomal DNA internal transcribed spacer (rDNA-ITS) sequence analysis, biological characteristics were using colony growth and slide methods.Among them, the results showed that the representative isolate was identified Fusarium solani.Effects of different cultures on mycelia growth were significant, optimal culture for mycelia growth was young-fruit decocted with liquid of jackfruit;effects of temperature,pH,carbon and nitrogen sources on mycelia growth and spore germination had significant differences, optimal temperature for mycelia growth was 28℃,fatal temperature for mycelia growth was 63 ℃,10 minutes;optimal temperature for spore germination was 25 ℃;pH7 was the optimal PH for mycelia growth;the optimal carbon source for mycelia growth was dextrose, ammonium nitrate was the optimal nitrogen sources for mycelia growth. This is the first report of root rot caused by F.solani on A rtocarpus heteophyllus Lam.
