Cloning and Bioinformatics Analysis of GPAT9 Gene in Rapeseed
GPAT9 is one of the important genes in fat metabolism of plants,but the research of this gene is not in-depth in Brassica napus.Studying the GPAT9 gene of Brassica napus,we designed a pair of primers,and according to the homologous comparsion of B.campestris,B.oleracea and A rabidopsis thaliana GPAT9 sequence,we cloned a full length CDs sequence that has 1 131 bp and that we named BnGPA T9.BnGPAT9 encodes a protein of 376 amino acids with the molecular weights of 43.25 kD,the instability index of 46.46 and the isoelectisc point of 8.97,and it encodes a protein of three TMHs without SignalP.The second structure of BnGPAT9 has 38.03% alpha helix,21.28% beta sheet,9.04% turn and 31.65% coil.Phylogenic tree analysis of BnGPAT9 indicated that BnGPA T9 was closely related to the GPAT9 in B.campestris,B.oleracea and Arabidopsis thaliana.BnGPAT9 possesses LPCAT1-like in Lysophospholipid acyltransferase superfamily and GPAT9 motifs.In terms of sequence comparsion,the analysis of physic-chemical property,subcellular localization and conserved domain,they showed that BnGPAT9 was Xiangyou 15 GPA T9 gene,and it should be the members of GPA Ts.
