马铃薯不同品种(系)遗传多样性的ISSR分析

本研究运用ISSR分子标记技术对30份马铃薯品种(系)材料进行多态性和聚类分析,分析各品种(系)间亲缘关系.结果表明:从56条ISSR引物中筛选出6条引物进行PCR扩增,共扩增出52条带,多态性条带为47个,多态性比率约为90.38%.通过UPGMA法对30份马铃薯品种(系)材料聚类分析显示,30份供试材料品种间遗传相似系数为0.40~0.95,平均相似系数为0.675;在相似性系数0.75附近,30份马铃薯品种(系)可分为6个大类,包括5大复合组和1个独立类.这些结果说明所选取的30个马铃薯品种(系)的遗传差异比较大,遗传多样性较丰富;该研究结果为马铃薯的遗传多样性利用及种质鉴定和杂交育种提供了有力的理论依据.
ISSR Analysis on Genetic Diversity of Different Varieties (Lines) in Solanum tuberosum L.
In this research,we used the ISSR molecular marker,in order to make polymorphism,cluster analysis and analyse the genetic relationships of 30 different varieties (lines) in Solanum tuberosum.The results showed that:After amplified 6 primers from 56 ISSR primer pools,total 52 DNA bands were amplified,of which 47 were polymorphic,and the polymorphic rate was 90.38%.Genetic similarity coefficient of 30 potato varieties (strains) changed from 0.40-0.95,with an average of 0.675.Cluster analysis results indicated that the potato varieties (strains) could be classified into 6 groups at 0.75 similarity level,including 5 composite groups and 1 independent group.These results indicated that 30 Solanum tuberosum varieties strains of the selected genetic differences,and the genetic diversity was abundant.In addition,the results of this study provided a strong basis for the utilization of genetic diversity,germplasm identification,and cross-breeding of Solanum tub erosum.
