基于ITS基因的部分兰属序列分析

本研究以4个兰属品种为材料,通过PCR扩增及构建系统发育树,分析4个兰属品种之间的亲缘关系.结果表明:(1)4个兰属品种的ITS序列长度在647~655 bp之间,其中ITS1的序列长度为242 bp,ITS2的序列长度为244 bp.(2)四个品种共存在175bp的变异位点,其中ITS1和ITS2的变异位点数较多,5.8S区域较为保守.(3)4个兰属品种的(G+C)含量在60.12%~68.21%之间,其中文山红柱兰的(G+C)含量最高,垂花兰的(G+C)含量最低.研究认为,ITS基因可以作为植物系统发育研究的标记,而且具有一定的灵敏度,为兰属植物的系统发育研究提供了较好的技术支持.
Sequence Analysis of Cymbidium Based on ITS Gene
Genetic relationship of 4 Cymbidium varieties was analyzed via PCR amplication and contruction of phylogenetic tree.Results showed:(1) Length of four varieties on ITS site were among 647 bp to 655 bp,the ITS 1 had 242 bp,and ITS2 had 244 bp.(2) 175 bp variation sites were existed in four varieties,and the ITS1 and ITS2 region had high variation.(3) Content of(G+C) were among 60.12% to 68.21%,C.wenshanense had the highest content,and C.cochleare got the opposite result.These results illustrate the ITS gene can be seen the marker to studyphylogenetic relationship,and has high sensitivity,could be offered exact support for Cymbidium phylogenetic study.
