Callus Induction and Plant Regeneration of Decaisnea insignis
In order to investigate the effects of various plant growth regulator on the induction of Adventitious bud and proliferation of Decaisnea insignis,immature seed were used as explants for tissue cultural experiment,and massive propagation and regeneration plants were obtained to establish a high-frequency regeneration system of Decaisnea insignis.The results showed that the optimum medium for callus induction of immature seeds was MS medium supplemented with 0.1 mg/L KT and 2.0 mg/L 2,4-D,and the induction rate reached to 92%.The optimum medium of adventitious shoot propagation was 1/2 MS medium supplemented with 0.5 mg/L NAA and 3.0 mg/L 6-BA,the reproduction coefficient reached to 14.49~19.32.The optimum Medium for Rooting was 1/2 MS medium supplemented with 0.6 mg/L NAA,and the rooting rate was 79.83%,the root system and small plants grew well.The complete regenerated plants were transplanted to the mixed matrix (humus:river sand =1:1) after domestication,and the survival rate could reached 90%.
