榆树叶形性状遗传变异与SSR标记关联分析

本试验以从张家口市黑龙山地区采集的40个榆树无性系为试验材料,对叶形性状和SSR分子标记进行关联分析.结果表明,在40个榆树无性系间,15个叶形性状均有着极显著变异;大多数叶形指标的重复力均在0.8以上.利用15个叶形性状对40个榆树无性系进行聚类分析,可分为4类,各无性系间遗传距离在0.578~9.309之间.以从17对引物中选出的13对多态性高的SSR引物对40个榆树无性系进行聚类分析,可完全分为4类,各无性系间的遗传距离在0.030~0.664之间.15个叶形性状与SSR分子标记达到极显著相关,相关系数为0.473 8 (p<0.001,n=780,rα=0.118).这说明,叶形性状与所选SSR位点关联水平较高.
Correlation Analysis of Elm Tree Leaf Characters Variation and the SSR Markers
This experiment collected 40 Elm clones from Zhangjiakou City Dragon Mountain Region as experimental material.In this research,we analyzed the leaf shape traits and SSR molecular markers.The results showed that,15 leaf traits had a significant variation between 40 Elm clones;Most the repeatability of the leaf shape index is above 0.8.Use 15 leaf shape index to 40 elm clones for clustering analysis,40 Elm clones could be divided into 4 kinds,the genetic distance of each clone was between 0.578~9.309.Use 13 pairs high polymorphic SSR primers selected from 17 primers to 40 elm clones for clustering analysis,the genetic distance of each clone was between 0.030~0.664.15 leaf traits and SSR molecular markers achieve significant correlation,the correlation coefficient was 0.473 8 (p<0.001,n=780,rα=0.118).This showed that leaf shape traits had high level associated with selected SSR loci.
