Genetic Diversity Analysis with Capillary Electrophoresis Detection with Fluorescent ISSR Markers on Foxtail Millet
Foxtail millet,originated from China,is an important crop with high genetic diversity.In recent years,many germplasm were developed,but germplasm resources are still limited.The genetic diversity of 30 foxtail millet germplasms was elevated by capillary electrophoresis detection with fluorescent ISSR markers,which has a higher degree of automation,accuracy and some other advantages.Eight ISSR primers were used to amplify 30 individuals.A total of 120 ISSR bands were obtained by 8 primers.Genetic similarity coefficients were changed from 0.89 to 0.93.The results showed that some individuals from different regions were clustered into the same group indicating that the relationships among these genotypes were close due to domestication and artificial breeding.This study could provide a theoretical base for classification and identification of millet varieties,scientific utilization of millet germplasm and molecular marker assisted breeding.
