Genetic Diversity Analysis of Manglietia conifera Dandy Germplasm by SSR Markers
In this study,8 SSR primer pairs with polymorphism were used to assess genetic diversity and relationship of 13 provenances of Manglietia conifera from Vietnam.The results showed that there were 133 polymorphic SSR loci detected by 8 pairs of SSR markers,with a mean of 16.63 and 5.47 per locus,the average observed and expected heterozygosities,Shannon information index and Nei's gene diversity were 0.64,0.71,1.86 and 0.74,respectively.The average number of effect alleles,observed and expected heterozygosities of 13 provenances were 5.47,0.60 and 0.42,respectively,Shannon information index ranged from 1.41~1.80,and Nei's gene diversity ranged from 0.59~0.75,the difference of genetic polymorphism level among provenances was obvious.The average value of genetic identities among provenances was 0.834,ranged from 0.655~0.967;the average value of genetic distance was 0.194,ranged from 0.034~0.423.According to the UPGMA cluster analysis,the 13 provenances could be obviously divided into four groups when genetic distance was 0.16,the phylogenetic relationships between provenance LC5 and other provenances were relatively distant,the classification results at the molecular genetic level were not completely consistent with the classification of the material source,there were no significant correlation between genetic and geographic distance among the M.conifera provenances.These results suggested that there is a high level of genetic polymorphism among M.conifera germplasms revealed by SSR markers.
