小麦TaROP2基因在生长发育中生物学功能的初步鉴定

植物ROP (Rho-like GTPases from plants)蛋白是小GTP结合蛋白Rho家族的成员之一,参与调控花粉管的极性伸长、根毛的发育、细胞骨架的重排、细胞发育与形态建成、细胞的内膜运输等.为探索ROP蛋白参与小麦生长发育过程中的作用,我们克隆了小麦TaROP2基因,利用生物信息学方法对TaROP2氨基酸序列中的保守结构域进行了分析.采用荧光定量方法,我们发现TaROP2基因在小麦幼苗分生组织中表达量相对较高.亚细胞定位分析结果表明,小麦TaROP2蛋白定位于细胞膜和细胞核.我们还构建了TaRO P2基因的植物过表达载体pGWB6-TaROP2,并获得拟南芥异源过表达目的蛋白的转基因植株.表型分析发现TaROP2基因参与拟南芥侧根形成和气孔发育.该研究为研究小麦TaROP2基因在生长发育中生物学功能,奠定了实验基础.
Preliminary Identification of Biological Function of Wheat TaROP2 Gene in Growth and Development
Plant ROP (Rho-like GTPases from plants) protein is one of the members of small GTP binding protein Rho family,which is involved in the regulation of pollen tube polarity elongation,root hair development,cell skeleton rearrangement,cell development and morphology,cell intima transport,etc.In order to explore the role of ROP protein in wheat growth and development,TaROP2 gene of wheat was cloned.The conserved domain of TaROP2 amino acid sequence was analyzed by bioinformatics.We found that the expression level of TaROP2 gene was relatively high in wheat seedling tissue by fluorescence quantitative analysis.Subcellular localization analysis indicated that TaROP2 protein was located in both cell membrane and nucleus.The plant overexpression vector pGWB6-TaROP2 was constructed and the transgenic plants for heterologous expression of A rabidopsis thaliana target proteins were obtained.Phenotypic analysis found that TaROP2 gene involved in lateral root formation and stomatal development.This study laid the experimental foundation for studying the biological function of wheat TaROP2 gene in growth and development.
