红果枸杞EST-SSR分子标记开发及应用

为了快速区分市场上不同品系的宁夏枸杞(Lycium barbarum L.),我们针对8种宁夏枸杞中进行了开发新型SSR引物,结果显示这些EST-SSR标记可用来调查分析枸杞的遗传结构.除此之外,运用SSR标记并进行聚类,表明供试的材料遗传多态性较高,8个枸杞品系拥有较高水平的多样性位点,其中宁杞1号和宁杞3号的遗传相似性最高,宁杞8号、宁杞3号和宁杞5号相似性系数为0.222 2,相似性最低,亲缘关系较远,因此这13对SSR引物可被用来快速鉴定宁夏枸杞的8个品系,为枸杞品种鉴定和保护,宁夏枸杞的道地性研究提供科学依据.
Development and Application of EST-SSR Molecular Marker for Lycium barbarum L.
In order to rapidly distinguish 8 different strains ofLycium barbarum,we collected 8 Ningxia Lycium barbarum to develop new SSR primers,and the results showed that these EST-SSR markers could be used to investigate and analyze the genetic structure ofL.barbarum.In addition,the SSR markers and clustering were used to show that the genetic polymorphisms of the tested materials were high,and 8 lines of Lycium barbarum had a high level of diversity loci,of which Ningqi 1 and Ningqi 3 had the highest genetic similarity.The similarity coefficient of Ningqi 8 and Ningqi 3 and 5 was 0.222 2,the similarity was the lowest and the phylogenetic relationship was far.Therefore,the 13 SSR primers could be used to identify the eight lines ofL.barbarum.For the identification and protection ofL.barbarum varieties,its scientific research might provide a scientific basis.
