Germination Characteristic of Rosa xanthina Pollen in vitro
In order to clear the pollen germination characteristic of Rosa xanthina, the effects of the concentrations of sucrose, H3BO4, CaCl2, NAA and GA, the cultivation temperature, collecting time and the storage method on pollen germination of Rosa xanthina were studied using pollen culture in vitro method. The results showed that cultivation temperature and the concentrations of sucrose, H3BO4, CaCl2, NAA and GA in the medium had significant influence on pollen germination, and the germination rate was different in different collecting time and different pollen storage methods in one day. The best germination effect was observed on 15% sucrose medium at 25℃, and the pollen germination rate reached 67.51%. When H3BO4 (150 mg/L), CaCl2 (20 mg/L), NAA (20 mg/L) and GA (20 mg/L) were added to the basic medium respectively, the better pollen germination rate was observed. The germination rates had no significant differences among the pollen that were stored at 4 different conditions of room temperature with flower or in vitro, and at 0℃~2℃ with flower or in vitro and cultivated after 5 h and 10 h. While at room temperature, the better effect was obtained when pollen was stored with flower than in vitro. However, at 0℃~2℃, the higher germination rate was observed when pollen was stored in vitro than with flower. The suitable time for pollen collection was from 10:00 to 14:00, and the highest germination rate was appeared when pollen was collected at 12:00. This experiment discussed the influence of different conditions on pollen germination and would provide basis for the cross breeding and theoretical research of Rosa xanthina.
