摘要
为了深入分析龙眼DlFNS蛋白的结构及其生物学功能,本研究从龙眼转录组数据(SRAaccession:SRP155595)中筛选和鉴定出一个DlFNS基因,并运用生物信息学方法对其进行分析.结果表明,DlFNS蛋白序列中含有530个氨基酸,分子式为C2731H4353N735O764S21,具有跨膜结构但不含信号肽,是亲水蛋白,主要定位在内质网中.二级结构预测结果显示,无规则卷曲和α-螺旋是DlFNS蛋白中主要的二级结构元件.利用SWISS-MODEL构建三级结构模型,并利用Ramachandran评估三级结构模型,结果表明构建的三级结构模型可靠.FNS蛋白的系统进化树分析表明DlFNS蛋白与阿月浑子FNS蛋白亲缘关系最近.本研究结果将为进一步研究龙眼DlFNS基因的功能及其在龙眼类黄酮合成中的分子调控机制提供理论依据.
Abstract
The aim of this study is to analyze the structure of the DlFNS protein identified from Dimocarpus lon-gan and its biological function.In this study,a flavone synthase(FNS)gene was screened and identified from D.lon-gan RNA-seq data(SRA accession:SRP155595).Multiple bioinformatics approaches were adopted to systemati-cally analyze the DlFNS gene.The results showed that the DlFNS protein translated from the DlFNS gene contains 530 amino acids,with its molecular formula being C2731H4353N735O764S21.It has a transmembrane structure but does not contain signal peptides.It was predicted to be a hydrophilic protein locating in endoplasmic reticulum.The secondary structure of DlFNS protein was analyzed,suggesting that DlFNS protein was mainly composed of ran-dom coil and α-helix.According to the evaluation by Ramachandran,the tertiary structure of DlFNS protein con-structed by SWISS-MODEL was reliable.The results of phylogenetic tree indicated that the DlFNS protein has a close relationship with a FNS protein identified from Pistacia vera L.The results of this study will facilitate the fur-ther study on the function of DlFNS gene and lay a basis for elucidating its role in terms of mediating the biosyn-thesis of flavonoid.
