Screening of Internal Reference Genes by Quantitative Real-time PCR in Oidium heveae under High Temperature Stress
In order to screen out the appropriate reference genes to study the gene expression of Oidium heveae at different growth stages under high temperature stress,seven candidate reference genes including EF1,GAPDH,CH,H3,PBS2,TUB and CU were determined by quantitative real-time PCR,and their expression stability was analyzed by △Ct method,GeNorm,NormFinder and Refindre.The gene expression stability order at conidial stage was EF1>H3>PBS2>TUB>GAPDH>CH>CU.The gene expression stability order were GAPDH>EF1>PBS2>CH>TUB>H3>CU at germination stage and EF1>GAPDH>PBS2>H3>TUB>CU>CH at infection stage.At hyphal growth stage and sporulation stage,the gene expression stability order were EF1>GAPDH>CH>H3>CU>TUB>PBS2 and EF1>GAPDH>H3>PBS2>CU>TUB>CH,respectively.In conclusion,the EF1 gene can be used as an ideal reference gene for the gene expression of O.heveae under high temperature stress.The results of this study provided internal refer-ence genes for the expression of related genes in O.heveae under high temperature stress.
