Cloning and Transformation of CIM Gene from Medicago truncatula
CIM(Cytokinin induced message)is a gene in the expansin-B2 family.Its main function is to affect the relaxation of plant cell walls,which affects plant growth and development as well as the response to adversity stress.In this study,Medicago truncatula R108 was used as the test material and CIM gene was isolated by PCR.The encoding region length of CIM gene was 828 bp and it could encode 275 amino acids.Bioinformatics analysis showed that the expansin proteins expressed by CIM of Medicago truncatula and expansin-B2 proteins of Cicer ari-etinum had high homology.Plant expression vector 35Spro∷CIM was constructed by DNA recombination technol-ogy,and transformed into alfalfa by Agrobacterium-mediated method,and 20 transgenic plants were successfully ob-tained.This study provides data reference and experimental materials for further exploring the function of CIM gene.
