蒺藜苜蓿CIM基因的克隆及转化

CIM(Cytokinin induced massage)是expansin-B2家族的基因,该基因的主要功能是影响植物细胞壁的松弛程度,从而影响植被由幼苗长成成熟植株的过程以及对各种环境胁迫的抵抗.本研究选取蒺藜苜蓿(Medicago truncatula)品种R108作为试验材料,采用PCR技术克隆出CIM基因.CIM基因的长度为828 bp,能够编码275个氨基酸.生物信息学分析显示:与蒺藜苜蓿CIM基因表达的expansin蛋白亲缘性相近的是鹰嘴豆(Cicer arietinum)expansin-B2蛋白.利用DNA重组技术构建植物表达载体35Spro∷CIM,然后通过农杆菌侵染的方法将35Spro∷CIM整合到紫花苜蓿叶片的基因组中,经过一段时间的组织培养,最终成功获得了 20株转基因植物.本试验为以后探究CIM基因对植物发育进程的作用给出了数据参考和试验材料.
Cloning and Transformation of CIM Gene from Medicago truncatula
CIM(Cytokinin induced message)is a gene in the expansin-B2 family.Its main function is to affect the relaxation of plant cell walls,which affects plant growth and development as well as the response to adversity stress.In this study,Medicago truncatula R108 was used as the test material and CIM gene was isolated by PCR.The encoding region length of CIM gene was 828 bp and it could encode 275 amino acids.Bioinformatics analysis showed that the expansin proteins expressed by CIM of Medicago truncatula and expansin-B2 proteins of Cicer ari-etinum had high homology.Plant expression vector 35Spro∷CIM was constructed by DNA recombination technol-ogy,and transformed into alfalfa by Agrobacterium-mediated method,and 20 transgenic plants were successfully ob-tained.This study provides data reference and experimental materials for further exploring the function of CIM gene.
