植物转化酶家族基因鉴定及序列进化分析

转化酶 CWIN(Cell wall invertase)、VIN(Vacuolar invertase)和 CIN(Cytoplasmic invertases)是蔗糖代谢途径中蔗糖转化成葡萄糖和果糖的关键酶,直接参与了植物蔗糖代谢的生物合成.为了系统梳理转化酶相关基因在植物基因组中的状况,实验室对其基因进行了全面的鉴定和进化分析,并重点考察植物在拟南芥和番茄中的表达.首先,实验室针对CWIN、VIN和CIN的基因序列,鉴定得到其保守结构域,在此基础上对相关基因进行了全面鉴定,从而构建了其进化树;其次,将拟南芥(Arabidopsis thaliana)和番茄(Salanum lycoper-sicum)CDS序列比对到各探针,从而从数据库中提取组织中的表达数据;最后,通过结构域鉴定、进化树和表达量的分析,得出CWIN、VIN和CIN在植物中的生物合成的机理.本研究为转化酶相关基因提供了基因信息,为植物中蔗糖代谢生物合成途径的作用机理及对蔗糖代谢影响相关基因进化机制的研究提供帮助.
Identification,Sequence Analysis and Evolutionary Inference of Plant Inver-tase Family Genes
Invertase CWIN(Cell wall invertase),VIN(Vacuolar invertase)and CIN(Cytoplasmic invertases)is the key enzyme in sucrose metabolic pathway to convert sucrose to glucose and fructose,which is directly in-volved in the biosynthesis of plant sucrose metabolism.In order to systematically comb the status of invertase re-lated genes in plant genomes,laboratory carried out a comprehensive identification and evolutionary analysis of its genes and focused on the plant expression in Arabidopsis thaliana and Solanum lycopersicum.Firstly,the laborato-ry identified the conserved domain of CWIN,VIN and CIN gene sequences,and on this basis comprehensively iden-tified related genes and constructed their evolutionary tree.Secondly,A rabidopsis thaliana and Solanum lycoper-sicum CDS sequences were aligned to each probe to extract tissue expression data from the database.Finally,through domain identification,evolutionary tree analysis and expression analysis,the biosynthesis mechanism of CWIN,VIN and CIN in plants was obtained.This study provides genetic information for invertase related genes and is helpful to study the mechanism of biosynthesis pathway of sucrose metabolism and the evolutionary mecha-nism of genes related to sucrose metabolism in plants.
