水稻脂肪氧化酶突变体功能标记开发与应用

稻谷在长期贮藏过程中会产生严重的陈化,主要原因是稻谷本身脂质的降解.在成熟的种胚中存在3种LOX同工酶,其总活性的80%~90%为LOX-3,该酶的缺失可以延缓稻谷的陈化变质.其活性缺失体(Daw Dam)可以降低贮藏期间稻谷不饱和脂肪酸的过氧化水平,减少包括己醛、戊醛和戊醇等令人不快的陈化气味的累积,遗传分析的研究则证实LOX-3的缺失由单隐性基因控制.通过测序确认水稻突变型(MT)lox3基因与野生型(WT)lox3基因相比,基因编码的mRNA再编码区第1 497个核苷酸发生了 G到A的置换突变,导致了翻译提前终止.利用该分子标记对部分水稻重要种质资源的lox3基因型进行了鉴定,并选育了三份LOX3缺失的水稻新品系.本研究设计出一种引物组合,其中外引物lox3-o-F、lox3-o-R分别位于突变位点的两侧,内引物lox3-i-F、lox3-i-R在突变位点上.利用该引物组合对水稻品种的基因组DNA进行PCR扩增,通过扩增产物带型即可区分lox3基因是野生型(WT)、突变缺失型和杂合型.该基因标记位于lox3编码区内部,与其他标记方法相比检测更准确,四引物的设计通过一次普通PCR即可完成反应,且带型差异较大,可使用廉价的琼脂糖凝胶电泳分辨带型.此次发明引物组合能够有效鉴别水稻耐储藏基因lox3基因型,使用一次普通PCR和琼脂糖凝胶电泳的方法对lox3进行基因分型,较其他检测方法相比,检测操作更简便、准确,成本更低.本标记的开发可为lox3基因型的高效利用提供技术参考,通过储藏实验对稻谷进行了耐储性评价,表明水稻种胚LOX-3与稻谷储藏特性关系密切.
Development and Application of Functional Markers of Rice Lipoxygenase Mutants
Rice was seriously aged during long-term storage,mainly due to the lipid degradation of rice itself.There were three LOX isozymes in mature seed embryos,and 80%~90%of the total activity is LOX-3.The ab-sence of this enzyme could delay the aging and deterioration of rice.Its active deletion body(Daw Dam)could re-duce the peroxidation level of unsaturated fatty acids in rice during storage and reduced the accumulation of un-pleasant aging odors including hexanal,glutaraldehyde and pentanol.Genetic analysis studies confirmed that the deletion of LOX-3was controlled by a single recessive gene.Sequencing confirmed that compared with the wild-type(WT)lox3 gene,the mutant(MT)lox3 gene encoded by the gene had a substitution mutation from G to A in nucleotide 1 497 of the mRNA recoding region,resulting in the early termination of translation.Using this molecular marker,lox3 genotypes of some important rice germplasm resources were identified,and three new rice lines with lox3 deletion were selected.A primer combination was designed in this study,in which the outer primers lox3-o-F and lox3-o-R were located on both sides of the mutation site,and the inner primers lox3-I-F and lox3-I-R were located on the mutation site.Using this primer combination,the genomic DNA of rice varieties was amplified by PCR.The lox3 gene was wild-type(WT),mutant deletion type and heterozygous type.The gene marker was lo-cated in the lox3 coding region,and the detection was more accurate than other marker methods.The design of four primers could complete the reaction through one ordinary PCR,and the band type difference was large.Cheap agarose gel electrophoresis could be used to distinguish the band type.The primer combination of the invention could effectively identify the lox3 genotype of rice storage tolerance gene,and genotypes lox3 by using one-time ordinary PCR and agarose gel electrophoresis.Compared with other detection methods,the detection operation was simpler,more accurate and lower cost.The development of this marker could provide technical reference for the efficient utilization of lox3 genotype.The storage tolerance of rice was evaluated through storage experiments,which showed that LOX-3 of rice embryo was closely related to the storage characteristics of rice.
