促丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)在植物生长、发育、抗逆、抗病和激素信号转导过程中起重要的调控作用.本研究以陆地棉的cDNA为模板克隆得到GhMA PKKKK3基因,其开放阅读框为1 998 bp,编码665个氨基酸残基.通过荧光定量PCR的方法分析GhMA PKKKK3基因在不同逆境诱导条件下以及不同棉花品种中的表达模式.结果表明,GhMA PKKKK3基因对高盐、干旱、低温和棉花黄萎病菌处理都有一定程度的响应,并且该基因受盐胁迫时调控的程度最强,其次是干旱胁迫与冷胁迫,黄萎病菌诱导时仅在1 h左右显著高于对照;该基因在不同品种棉花根部组织中的表达量低于茎部和叶部组织,在不同黄萎病抗性的品种中GhMA PKKKK3基因表达量有差异但没有显著规律.本研究结果为棉花抗逆相关基因的筛选及进一步研究GhMA PKKKK3基因的功能提供了重要的线索.
Cloning and Expression Analysis of GhMA PKKKK3 Gene in Gossypium hirsutum L.
Mitogen-activated protein kinase(MAPK)plays an important regulatory role in plant growth,develop-ment,stress resistance,disease resistance and hormone signal transduction.In this study,the GhMA PKKKK3 gene was cloned using the cDNA of Gossypium hirsutum as a template.Its open reading frame is 1 998 bp and encodes 665 amino acid residues.The expression patterns of GhMA PKKKK3 gene under different stress induction condi-tions and different cotton varieties were analyzed by RT-qPCR.The results showed that the GhMA PKKKK3 gene had a certain degree of response to high salt,drought,low temperature and Verticillium dahliae treatment,and the gene was regulated most strongly under salt stress,followed by drought stress and cold stress;when induced by V.dahliae,it was only significantly higher than the control at about 1 h.The expression of this gene in the root tissues of different varieties of cotton was lower than that of the stem and leaf tissues.GhMA PKKKK3 gene expression dif-fered among varieties with different Verticillium wilt resistance but there was no significant pattern.The results of this study provide important clues for the screening of cotton resistance-related genes and further study of the func-tion of GhMA PKKKK3 gene.
