抽穗期是水稻(Oryzasativa)的重要农艺性状,抽穗期的长短决定了水稻生育期的长短,合适的抽穗期对于维持水稻的稳产高产至关重要.Ehd1(Early heading date 1)是水稻中特有的抽穗期调控基因,对于调控粳稻在短日照条件下的抽穗期发挥着重要作用.'日本晴'、'越光'等粳稻品种含有功能正常的Ehd1基因,在短日照条件下抽穗期提前,'台中65'等粳稻品种的Ehd1基因在第三外显子中存在一个G到A的碱基突变,该碱基突变引起甘氨酸到精氨酸的置换,导致EHD1蛋白功能减弱,最终造成'台中65'等粳稻品种在短日照条件下抽穗期延迟的表型,Ehd1是短日照条件下对粳稻抽穗期进行遗传改良的重要靶标.本研究根据Ehd1第三外显子的功能性变异位点,应用扩增受阻突变体系(amplification refractory mutation system,ARMS)的原理,开发四引物分子标记Ehd1-ARMS,利用ARMS PCR技术对Ehd1的不同等位基因进行特异性扩增,为快速、准确地对Ehd1进行基因分型,以及优质粳稻在短日照条件下的遗传改良等提供帮助.
Detection of Ehd1 Gene for Heading Date by Tetra-Primer Amplification Refractory Mutation System PCR in Rice
Heading date is an important agronomic trait in rice(Oryza sativa),the length of heading period has an significant effect on rice growth period,an appropriate heading date is crucial for maintaining high yield in rice.Ehd1 is a unique gene that can regulate the heading date in rice,especially in regulating the heading date of japonica rice under short-day conditions.Most japonica rice varieties such as'Nipponbare'and'Koshihikari'contain a functional Ehd1 gene,which can lead to early heading under short-day conditions.The Ehd1 gene in a few japonica rice varieties,such as'Taichung65',exhibits a G to A nucleotide mutation in the third exon.This single nucleotide mutation causes a substitution of glycine to arginine,which can lead to a weakened function of EHD1 protein,ultimately resulting in a delayed heading phenotype in japonic a rice under short-day conditions.The Ehd1 gene is an important target for genetic improvement of heading date in japonica rice under short-day conditions.This study developed a tetra-primer molecular marker Ehd1-ARMS based on the Ehd1 functional variation sites and the principle of amplification refractory mutation system(ARMS).The molecular marker Ehd1-ARMS can specifically amplify different Ehd1 alleles,providing assistance for rapid and accurate genotyping of Ehd1 and heading date genetic improvement of Japonica rice.
