Screening and Identification of ERF Transcription Factors Related to Water-logging Stress in Petunia
ERF transcription factors are a type of plant-specific transcription factors and belong to the AP2/ERF transcription factor family,regulate a variety of biological functions and play an important role in the response of plants to biotic and abiotic stresses.This research chose PhERF2-overexpressing transgenic Petunia line(I)and Ph-ERF2-RNAi transgenic Petunia line(4B)as the experimental materials,compared with wild type(WT).Differen-tially expressed gene data in Petunia exposed to 0 and 3 h of waterlogging treatment were obtained,which based on transcriptome sequencing technology(RNA-Seq).Bioinformatics methods were used to analyze the conserved motifs and the physicochemical properties of the proteins,constructed phylogenetic tree,modeled the advanced structure of the proteins,predicted the phosphorylation modification sites and analyze the subcellular localization.The re-sults showed that 35 ERF transcription factors related to waterlogging stress were screened out,and they were members of AP2 subfamily.Phylogenetic analysis with Arabidopsis and rice genes indicated that Petunia and Ara-bidopsis shared high homology.Bioinformatics analysis showed that waterlogging stress-related ERF transcription factors in Petunia were rich in acidic amino acids,with high thermal stability,and they all belong to the hy-drophilic proteins.Their protein secondary structures were mainly composed of random curls.The differences in protein tertiary structure was not obvious,consisting of three conservative motifs.Most of the NAC proteins were located in the nucleus,containing potentially serine(Ser),threonine(Thr),and tyrosine(Tyr)phosphorylation sites.This study provids a basis for further functional exploration of ERF genes related to waterlogging stress in Petunia.
