Fingerprint Construction and Genetic Diversity Analysis Based on SSR Markers for'XinshiK28'
DNA fingerprinting was used to distinguish'XinshiK28'from other materials with similar genetic back-grounds to ensure the authenticity of this variety.In this study,30 pairs of core SSR primers were used for molecular detection of'XinshiK28'and its parent materials and 42 cotton varieties in Xinjiang.In the meantime,the DNA fingerprint and QR code information of'Xinshi K'series varieties were constructed.These results showed that the 30 pairs of polymorphic SSR primers amplified 96 alleles,and each pair of primers could amplify 2~7 sites,with an average of 3.2 alleles.Cluster analysis showed that 42 cotton materials could be grouped into four categories when the Jaccard coefficient was 0.71,which implies that the genetic differences between cotton materials were smaller and the genetic relationship was closer.The 30 pairs of SSR core primers can provide technical support for the authenticity identification of'XinshiK28',and provide a reference for the cultivar registration and the plant variety protection.
