SNP Markers Development and Genetic Diversity Analysis of Schima su-perba
Develop the SNP locus of Schima superba,and based on SNP molecular marker technology,reveal the genetic diversity and variation of Schima superba germplasm resources at the DNA molecular level.In this study,15 populations(provenances)in Guangdong,Fujian,and Jiangxi provinces were selected as the research materials,and a total of 179 Schima superba individual plants were selected for Restriction-site associated DNA sequencing(RAD-seq).Finally,16 383 SNP polymorphic loci were obtained,and use bioinformatics analysis software to ana-lyze genetic diversity and genetic structure 179 individual Schima superba plants were analyzed,and the average observed heterozygosity(Ho)was 0.074 1;the average expected heterozygosity(He)was 0.293.Analyzing 15 Schi-ma superba populations,the average observed heterozygosity(Ho)is 0.074;the average expected heterozygosity(He)is 0.247;the genetic distance(d)between populations is distributed from 0.298 to 0.396,and the average is 0.350;the average polymorphic loci percentage(PPL)was 57.93%;the polymorphic information content(PIC)of each population ranged from 0.301 to 0.364,with an average of 0.335.All 15 Schima superba populations showed moderate polymorphism.Through genetic structure analysis,principal component analysis and cluster analysis of 15 Schima superba populations,15 Schima superba provenances were divided into four categories.There are abun-dant SNP sites in the Schima superba genome,and the polymorphism is moderate to upper,and it has a relatively rich genetic diversity.SNP molecular markers have potential value in the selection and breeding of Schima super-ba,increasing genetic gain and shortening the breeding cycle.
