The Protein Identification and Analysis of the Leaves from Hainan Bruguiera gymnorhiza under Pb2+and Cd2+Stresses
Bruguiera gymnorhiza is one of the medicinal plants of Hainan.The two dimensional electrophoresis technique based on BPP+phenol extraction was carried out to obtain the plant protein fingerprint of Bruguiera gymnorhiza leaves for the purpose of further research and development of the leaves from Bruguiera gymnorhiza.Some high expression proteins have been screened and analyzed by using the Image Master software.Then the enzymatic hydrolysis of these highly expressed proteins and their protein names were identified by MALDI-TOF-MS(matrix-assisted laser desorption ionization time-of-flight mass spectrometry)as well as the functions of these highly expressed proteins were be preliminarily analyzed.In addition,the differential proteins of Bruguiera gymnorhiza leaves under the stress of Pd2+and Cd2+were separated,extracted and identified respectively.The results indicated that there were sixty highly expressed proteins in the leaves of Hainan Bruguiera gymnorhiza and forty-two proteins or proteases including epimerase,DNA ligase were identified by biomass spectrometry,which involved in multiple functions of Bruguiera gymnorhiza leaves such as growth,development,heredity and plant resistance.There were 4 and 6 differential proteins under the stress of Pd2+and Cd2+,respectively,which are associated with plant growth,heredity and high efficiency of photosynthesis.The study not only provides the theory foundation for the further exploitation and utilization of the plant proteins in the leaves of Hainan Bruguiera gymnorhiza,but also has some theoretical and practical significance for finding the key proteins associated with the stress of Pd2+and Cd2+,understanding the ecosystem service and ecological benefits of Hainan Island.
