Genetic Analysis and Mapping of Wrinkle Leaf Gene wl in Melon
Melon leaf is the main organ of photosynthesis,and its morphology has an effect on the content of soluble solids(sugar content)in melon fruit.In order to genetic analysis and mapping wrinkle leaf gene wl in melon and the relationship between the leaf shape traits and fruit sugar,'Xizhou honey25'(X25)(wrinkled leaf)was the female parent and'Jiashi'(flat leaf)was the male parent to establish F2 and F2BC1 populations in this study.The separation ratio of wrinkled and flat plants in the two populations was measured by x2 test to analyze the inheritance of wrin-kled leaf genes.The results showed that the wrinkled leaf parent'Xizhou honey25'(X25)crossed with the flat leaf parent'Jiashi'(JS)to obtain the F1 generation of flat leaves,which indicate that wrinkled leaf to flat leaf was a recessive inheritance.Simple sequence repeat(SSR)molecular markers combined with bulk segregant analysis(BSA)method were used to construct DNA pools of wrinkles and flat leaves,and 396 SSR molecular markers across the whole genome were used to screen polymorphism markers.Six polymorphic SSR markers were found at LG7(CMGA165 and GCM344),LG8(ECM200 and CMACC146)and LG 10(ECM232 and CMCT134B),respec-tively.The F2 and F2BC1 genotypes were detected and the wrinkle leaf gene wl of melon was linked analysis and mapped.Finally,a major QTL locus of wrinkle leaf gene wl,which was located between marker CMCT134B and SSR2441(LOD>3),physical distance is 4.4 Mb.The data obtained in the field were analyzed for wrinkled leaf cha-racter and soluble solid(sugar content)data.Descriptive results of sugar content determination showed that the sugar content of wrinkled leaf character was significantly lower than that of flat leaf character,and there were significant differences between the two phenotypes.This study provides a reference for fine mapping and cloning of wrinkled leaf traits in melon.
