Development and Application of Arecanut InDel Markers Based on Resequencing
Arecanut(Areca catechu L.)is an important tropical economic crop in Hainan Province,the whole genome sequence of cultivated arecanut'Reyanyihao'has been released,that provides research basis for the development of InDel markers.In this study,we had compared the sequences of a long oval-shaped resource to reference genome,forty two InDel loci on chromosome 1 were chosen and designed primers,16 polymorphic InDel markers with clean bands were screened out,these 16 polymorphic InDel markers were further used for genetic diversity evaluation in 24 arecanut accessions,the polymorphic information content(PIC)value for each InDel marker range from 0.115 0 to 0.414 5,with an average value of 0.325 8,population cluster analysis revealed there are three groups existed in these accessions with the genetic similarity coefficient of 0.64,one ovate fruit from Yunnan was clustered into group Ⅲ,two resources from Taiwan and three materials from Sri Lanka were clustered into a branch,and most of the materials were clustered into group Ⅱ,the result indicated the resources collected and imported from foreign country had broadened the genetic basis of arecanut in Hainan.The InDel markers developed in this study can effectively reveal the relationships among germplasm resources based on the genetic distance,it will be effectively used in genetic linkage map construction and genome-wide association study.
