Cloning and Bioinformatics Analysis of OsIRT1 Gene from Rice(Oryza sati-va L.)'Yixiangyou 2115'
Iron-regulated transporter 1(IRT1)is the main root transporter for plant to absorb iron from soil,it is also the main way for toxic metals such as cadmium to enter plants.The IRT1 gene of iron-rich rice(Oryza sativa L.)was cloned to provide reference for studying the function of the gene in the process of iron transportation in plants.In this study,the IRT1 cDNA sequence amplified from'Yixiangyou 2115'rice plant by reverse transcription PCR(RT-PCR),and designated as OsIRT1-YXY2115.Sequence analysis revealed that the coding region cDNA of OsIRT1-YXY2115 was 1 125 bp,which encoding 374 amino acids,and it had 9 membrane-spanning domains,with a predicted molecular mass of 39.06 kD,a theoretical isoelectric point(pI)of 8.89,and the instability index in the solution was 45.73,it is an unstable protein belonging to the zinc iron transporter(ZRT/IRT-like protein,Zip)su-perfamily.The OsIRT1-YXY2115 amino acids showed high identity with the reported OsIRT1 gene of rice(AB070226.1)by NCBI homology comparison analysis.The sub-cellular localization of this protein was predicted to be the plasma membrane.The cloning of the OsIRT1-YXY2115 gene and preliminarily predicting its structure,nature and functions,provide a reference and theoretical basis for studying the biological function of the OsIRT1 gene and the molecular mechanisms underlying iron deficiency responses in rice.
