Cloning and Expression Pattern Analysis of MaGA2ox1 Gene in Brazilian Banana and Super Dwarf Banana
In order to cultivate lodging-resistant dwarf bananas,this study took Brazil banana(Musa spp.Baxi,BX)and super dwarf cavendish(SDC),which were commonly cultivated as materials,cloned the GA 2ox gene,flu-orescence quantitative expression and tobacco subcellular localization analysis.The cDNA length of this gene is 990 bp,encoding 329 amino acids,and the amino acid sequence of its encoded protein is completely consistent in the two materials.The molecular weight of the protein is 36.4 kD,the isoelectric point is 6.27,and there is no transmembrane region and signal peptide.The protein sequence synthesized by MaGA2ox1 gene is most closely re-lated to coconut,rice and Arabidopsis.The gene is localized in cell membrane and nucleus.At the five-leaf stage,the expression of MaGA 2ox1 gene in roots,leaves and heart leaves of SDC was significantly lower than that of Brazil banana.At the ten-leaf stage,the expression of MaGA 2ox1 gene in the heart leaves of SDC was significantly lower than that of Brazil banana.The total tissue expression of MaGA2ox1 gene in banana was higher than that in SDC.In this study,by analyzing the subcellular localization and expression pattern of MaGA2ox1 gene in Brazil banana and SDC,it provides a reference for studying the regulation of GA synthesis and the role of GA signal transduction during banana development,and the breeding of dwarf banana varieties.
