Gene Cloning and Expression Characterization of Five PaMYBs in Populus alba×P.berolinensis
MYB transcription factor is one of the important transcription factors in plants.It not only participates in the synthesis of flavonoids and anthocyanins,but also plays an important role in the process of abiotic stress re-sistance.In this study,five R2R3-MYB transcription factors(PaMYB116,PaMYB117,PaMYB118,PaMYB119,Pa-MYB1 20)were cloned from leaves and inflorescence of Populus alba.Analysis showed that the ORFs of the five PaMYBs were 789,777,822,825 and 750 bp,respectively.They were homologous to Arabidopsis AtMYB113 and parahomologous to anthocyanin synthesis.RT-qPCR showed that the expression of PaMYB116 was the highest in inflorescence,PaMYB118 and PaMYB120 were the highest in stems,and PaMYB117 was the lowest in all tissues.Five PaMYBs promoter sequences were found in the genome data of Populus alba and their cis-acting elements were predicted respectively.According to the predicted results,the Populus alba was treated with light and ABA respectively.The results of RT-qPCR showed that the five PaMYBs genes could be induced to express by light,among which PaMYB116,PaMYB117,PaMYB118 and PaMYB119 genes increased rapidly after dark treated for 3 days and then exposed to light for an hour.The relative expression of PaMYB120 was the highest at 48 h of light exposure;the relative expression levels of 5 PaMYBs genes were decreased under 200 μmol/L ABA treatment.The results provide a reference for revealing the MYB gene of Populus alba.
