聚合Wx-mp、fgr和Pi-ta、Pi-b基因选育优质粳稻新品种

以同时含有低直链淀粉含量基因Wx-mp、香味fgr 基因、稻瘟病抗性基因Pi-b的优良食味粳稻中间品系'宁7020'为母本,携带稻瘟病抗性基因Pi-ta的江苏抗病、高产粳稻品种'扬粳9538'为父本,配置杂交组合进行聚合育种.利用3套自主研发的PCR检测体系稻瘟病Pi-ta和Pi-b的多重PCR体系、Wx-mp基因的四引物扩增受阻突变体系PCR检测技术、香味fgr基因的InDel标记检测技术,分别在不同的世代进行目标基因跟踪,结合抗性、香味和籽粒胚乳外观表型鉴定,成功地将上述4个基因聚合于一体,选育出优质、高产、多抗、早熟的水稻新品系'南粳58'.本研究为选育多抗、优质水稻新品种提供了一种简单、快捷的选择技术,同时也为水稻抗病、优质育种提供新的种质资源.
The Breeding of New Japonica Rice Varieties with Good Quality by Pyra-miding Wx-mp,fgr,Pi-ta and Pi-b Genes
In this study,the high yield rice line'Ning7020'contained blast resistance gene Pi-b,low-amylose con-tent gene Wx-mp and fragrance gene fgr were used as receipt to be crossed with the rice variety'Yangjing 9538',which carries the blast resistance gene Pi-ta.Self-designed multiple PCR system to detect Pi-ta and Pi-b and te-tra-primer amplification refractory mutation PCR system to detect Wx-mp and InDel marker for fgr were used to detect the target loci in separation generation.Combining with resistance identification,fragrance characterization and classification of the grain endosperm appearance,a new japonica rice line'Nanjing58'with excellent agro-nomic traits was bred by multi-generational selection on resistance,fragrance and endosperm appearance.This study provides a simple and fast selection technique for breeding new varieties of multi-resistant and high-quality rice,and also provides new germplasm resources for rice disease resistance and high-quality breeding.
