Histochemical Observation of Anthers Development in Castanea mollissima
The male flowers of Castanea mollissima at different developmental stages were used as experimental materials to observe the accumulation and distribution of starch,polysaccharide and protein in the anther develop-ment of C.mollissima by histochemical method.The results showed the distribution of accumulation and tran-slocation of nutrients during the anther development of C.mollissima had displayed certain temporal and spatial characteristics.At stage of stamen primordium stage,a lot of polysaccharides were accumulated in the differen-tiation of sepal tissues.At stage of microspore mother cells,anther wall began to distribution of polysaccharides.The meiosis was cytoplasmic simultaneous type,at same time,starches were appeared in anther wall tissue,and proteins were contained in tapetum cells at stage of meiosis.At stage of tapetal,there were starches and poly-saccharides in anther wall and connective tissue increased,and proteins in tapetal cells was increased.At stage of early microspore,starches grains appeared in microspores,and tapetal cells accumulate proteins gradually in-creased.At stage of bicellular pollen,tapetal were only remnant and the cells began to accumulate large amounts of starches,polysaccharides and proteins.The mature pollen grains were 2-celled type,belonged to starch type pollen,and a large number of starches and a small amount of polysaccharides and proteins were accumulated in the pollen as nutrients.
