Effects of Stratification Conditions and Hormone Treatments on Seed Post-ripening of Epimedium koreanum Nakai
In order to investigate the mechanism of seed post-ripening in Epimedium koreanum Nakai and to solve the problem of seed breeding,seed post-ripening conditions under natural stratification and low temperature stratification were compared and the concentrations of hormones promoting embryo growth were further screened by using six to seven percentage mature seeds.The results showed that:under natural stratification(A),the seeds were first finished post-ripening,the shortest post-ripening time was 80 days;when stratification reached 120 days,the embryo length was 2.703 3 mm,the embryo width was 0.76 mm and the embryo rate was 70.33%,which was significantly better than low temperature stratification;under natural stratification the best concentration of gibberellin(GA3)was 400 mg/L,and the embryo rate reached 88.23%after 120 days of stratification.Under low temperature stratification,none of the exogenous hormones could obviously promote the development of seed embryos.In conclusion,the post-ripening process of seed embryos could be finished successfully after 120 days of natural stratification,and the effect of GA3 400 mg/L is better,this study provided a theoretical basis for post-ripeningand seed germination mechanism of Epimedium koreanum Nakai.
