Regulation and Analysis of Sucrose on Tuber Formation and DamiR156/DaSPL in Dioscorea alata
In order to explore the effects of sucrose on tuber formation of different genotypes of Dioscorea alata(great yam),as well as the regulatory effect of DamiR156 and its target gene DaSPL in the development of tubers,this study treated Da40,Da70,and Da207 of great yam with different sucrose concentrations,and the results showed that high concentrations of sucrose could promote tuber formation of Da40 and Da70.When the sucrose concentration was 90 g/L,the tuberculosis formation time of Da40 was 22 days earlier,and Da70 was 40 days earli-er.The relative expression of DamiR156 and its target gene DaSPL6 of Da70 under different sucrose concentration treatments was determined by RT-qPCR.The results showed that at 30 days,the expression of DamiR156 was high est at 70 g/L,and at 60 days(the early stage of tuber formation),the expression of DamiR156 was the highest and DaS-PL6 was the lowest at a sucrose concentration of 0 g/L.Under the treatment of 6 different sucrose concentrations,the expression of DaSPL6 showed a trend of first rising and then decreasing,and DaSPL6 was the highest expres-sion at 70 g/L.The results showed that exogenous sucrose could promote the growth of great yam and induce early tuber formation while regulating the expression of DamiR156/DaSPL6 and providing a theoretical basis for the sub-sequent study of the molecular mechanism of sucrose regulating the tuber formation of great yam.
