基于SSR分子标记的燕麦遗传多样性分析与异交率估算

通过SSR分析了 9份燕麦种质材料的遗传多样性,研究同一小穗上种子萌发形成的子株间的遗传差异,并估算了材料的异交率.引物共扩增出多态位点81个,平均多态信息含量(PIC)为0.485 2.各材料间、同一材料母株间、同一小穗上种子形成的子株间平均遗传相似性系数分别为0.688 0、0.879 6、0.964 0.在遗传相似性系数为0.80处,可将供试燕麦材料划分为9个类群.本研究首次在国内开展燕麦异交率的估测,通过SSR分子标记估算出9份供试燕麦材料的异交率范围是1.39%~6.11%,平均为3.79%.该结论证明燕麦是以自交为主并兼有一定比例异交的自花授粉植物.本研究结果为燕麦品种保纯以及杂交育种研究提供了重要参考.
Genetic Diversity Analysis and Outcrossing Rate Estimation of Oat Based on SSR Molecular Markers
By using SSR method,the genetic diversity of 9 oat materials and the genetic differences among the offsprings on the same spikelet were analyzed.Then outcrossig rate of each material was estimated.The amplified polymorphic loci were 81,and the average polymorphic information content(PIC)was 0.485 2.Among the ma-terials,the mother plant from the same material,the offsprings from the same spikelet,the average coefficients were respectively 0.688 0,0.879 6 and 0.964 0.All materials were divided into 9 groups when the coefficient was 0.80.In this study,the outcrossing rate of oat was measured firstly in China.Based on SSR,the outcrossing rate of 9 oat materials tested range from 1.39%to 6.11%,the average is 3.79%.It was proved that oat is a self-pollinated plant with a certain proportion of outcrossing.The results will provide important references for maintaining the pu-rity of oats varieties and the research on cross breeding.
