Genetic Diversity Analysis of Wild Populations of Primula maximowiczii Based on AFLP Markers
In order to provide the basis for the protection and development of wild Primula maximowiczii germ-plasm resources,seven typical wild populations of P.maximowiczii were selected to evaluate the genetic structure and genetic diversity by AFLP molecular marker.Six pairs of primers with high polymorphism were screened and 161 polymorphic loci were obtained.The higher percentage of polymorphic loci in WTS and LS population were 97.44%and 92.31%,respectively.The genetic diversity of intra-population accounted for 86.20%of the total ge-netic diversity,while inter-population accounted for 13.80%.The inter-population genetic differentiation was small(Gst=0.114).Analysis of molecular variance showed that there were significant differences between inter-popula-tion and intra-population(P<0.001),and the genetic variation of P.maximowiczii mainly came from intra-population.Nei's unbiased estimation and UPGMA cluster analysis showed that the seven populations were clustered from south to north according to geographical location,and the populations of HTS and LS had the smallest genetic dis-tance and the largest genetic consistency.The results verified the phenotypic diversity of P.maximowiczii popula-tions,provided references for the development and breeding of P.maximowiczii with high ornamental value,and provided guidance for the measures about the protection of wild P.maximowiczii resources.
