Cloning and Bioinformatics Analysis of EPSPS Gene from Rosa chinensis
Rose is a common flower in landscaping.5-enol acetonyl shikimic acid-3-phosphate synthase(EP-SPS)is a key synthase in the shikimic acid synthesis pathway and a target enzyme for the herbicide glyphosate.In order to explore the structure and function of rose EPSPS gene,this study used the genomic information of rose to design specific primers to clone EPSPS gene of rose,named RcEPSPS.The ORF of the RcEPSPS gene cloned by RT-PCR was 1 566 BP,encoding 521 amino acids,with a molecular weight of 55.19 kD,an isoelectric point of 7.5 and a hydrophilic coefficient of 0.009.The preliminary study showed that the rcEPSPS protein belongs to type ⅠEPSPS,which is highly homologous with the EPSPS protein sequence of wild strawberry of Rosaceae.The results can provide a theoretical basis for the subsequent use of resistance genes.
