Characteristics Analysis of the Chloroplast Genome of Cocculus laurifolius
In order to explore the characteristics of chloroplast genome of Cocculus laurifolius and its phylogenet-ic relationship with chloroplast genome of the same family.The Illumina HiSeq 4000 platform was used to se-quence,assemble and annotate Cocculus laurifolius,and its sequence characteristics,codon preference,sequence repeats,simple sequence repeats and phylogeny were analyzed.The results showed that the chloroplast genome of Cocculus laurifolius showed a typical tetrad structure,with a total length of 157 961 bp,including a large single copy region of 89 173 bp,a small single copy region of 20 198 bp and a reverse complementary repeat region of 24 295 bp;a total of 129 genes were detected,including 85 protein coding genes,36 tRNA genes and 8 rRNA genes.Codon preference analysis showed that leucine(Leu)had the largest number of codons(4 920),while cys-teine(Cys)had the least number of codons(568).A total of 26 long repeats and 65 SSR loci were detected from the chloroplast genome of Cocculus laurifolius.Phylogeny analysis showed that Stephania tetrandra and Cocculus lau-rifolius were clustered into one branch,and the relationship was the closest.This study provides scientific basis for molecular markers,species identification,kinship analysis and genetic diversity analysis of Tetrandridae and Stephania.
