SRAP Analysis of Genetic Diversity in Different Populations of Amygdalus mongolica
In this study,five different populations of A mygdalus mongolica were taken as the material research ob-ject,and the population of Prunus mongolica with the same genus and related species as the control.The genetic di-versity of rare and endangered medicinal plant Amygdalus mongolica was studied by SRAP molecular marker method,so as to provide a scientific basis for the rational protection and sustainable utilization of Amygdalus mon-golica resources.Using 12 pairs of SRAP primers screened,60 samples were amplified through the optimized reac-tion system.A total of 171 clear and repeatable bands were amplified,of which 166 were polymorphic,accounting for 97.08%.At the species level,Nei's gene diversity(h)was 0.371 1 and Shannon information index(I)was 0.546 8;The coefficient of genetic differentiation(Gst)among populations was 0.596 4.The results showed that the genet-ic variation of Amygdalus mongolica population mainly existed among populations,and the degree of genetic dif-ferentiation among populations was greater than that within populations.Through comprehensive evaluation,norikong had high genetic diversity and should be protected first.
