EMS诱变对藜麦种子萌发及幼苗生理特性的影响

为筛选藜麦种子甲基磺酸乙酯(EMS)诱变的最佳处理条件,本试验以'忻藜1号'为材料,采用培养皿萌发与营养土育苗的方法,研究了不同EMS浓度(0,1.4%,1.6%,1.8%,2.0%)和不同时间(50,100,150,200 min)诱变处理下,藜麦种子发芽率、出苗率及幼苗脯氨酸含量、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性和过氧化物酶(POD)活性等生理指标的变化.结果表明:(1)藜麦的发芽率和出苗率随着EMS浓度的升高和处理时间的增加而下降,且差异显著.当EMS浓度为1.8%、处理时间为200 min时,藜麦发芽率和出苗率最接近半致死量.(2)藜麦叶片中脯氨酸(Pro)含量,随EMS浓度和处理时间的增加而先升后降;随着EMS浓度的增加与处理时间的延长,藜麦叶片中MDA含量总体上均有不同程度的增加;当EMS浓度为2.0%时,藜麦叶片中MDA含量随时间的增加而呈现出先升后降的趋势,且显著高于对照.(3)随EMS浓度和处理时间的增加,藜麦叶片中超氧化物歧化酶(SOD)、过氧化物酶(POD)活性总体呈先升后降的趋势.由此可得,EMS诱变藜麦最佳浓度与处理时间为1.8%、200 min.本研究为藜麦种质资源的创新及突变体库的构建奠定基础.
Effects of EMS Mutagenesis on Seed Germination and Physiological Char-acteristics of Quinoa Seedlings
In order to screen the optimal treatment conditions for ethyl methanesulfonate(EMS)mutagenesis of quinoa seeds,this experiment was conducted to study the changes in germination rate,seedling emergence rate,and physiological indexes[such as proline content,malondialdehyde(MDA)content,superoxide dismutase(SOD)activity,and peroxidase(POD)activity)]of quinoa seeds under different EMS concentrations(0,1.4%,1.6%,1.8%,2.0%)and durations of treatments(50,100,150,200 min).The'XINLI1'variety was used as the experimental material,and Petri dish germination and nutrient soil seedling nursery methods were employed.The results showed that:(1)The germination rate and seedling emergence rate of quinoa decreased significantly with the increase of EMS concentration and treatment time.When the EMS concentration was 1.8%and the treatment time was 200 min,the germination and seedling emergence rates of quinoa were closest to the medial lethal dose.(2)The proline(Pro)content in quinoa leaves initially increased and then decreased with the increase of EMS concentration and treatment time.MDA content in quinoa leaves generally increased to varying degrees with increasing of EMS con-centration and treatment time.When the EMS concentration was 2.0%,MDA content in quinoa leaves showed a tendency to increase and then decrease with increasing treatment time,and it was significantly higher than that of the control.(3)The activities of superoxide dismutase(SOD)and peroxidase(POD)in quinoa leaves generally increased and then decreased with increasing EMS concentration and treatment time.Overall,the optimal concen-tration and treatment time for EMS mutagenesis of quinoa were determined to be 1.8%and 200 min,respec-tively.This study provides a foundation for innovation in quinoa germplasm resources and the establishment of mutant libraries.
